salting-out procedure
نویسندگان
چکیده
DNA from archival Papanicolaou stained and unstained cytological smears was successfully isolated using a simple, rapid and inexpensive salting-out procedure. The quality of DNA was controlled by polymerase chain reaction (PCR) amplification of segments of the human P-globin, human P-actin and human papillomavirus LI genes. Only negligible differences in amplification efficiency were observed between DNA isolated from stained and unstained smears. The salting-out procedure is a more rapid method for the isolation of DNA than phenolchloroform extraction and may be used in instances where fresh or cryopreserved clinical specimens are not available. متن کاملQualification Study of Two Genomic DNA Extraction Methods in Different Clinical Samples
INTRODUCTION The purity of genomic DNA (gDNA) extracted from different clinical specimens optimizes sensitivity of polymerase chain reaction (PCR) assays. This study attempted to compare two different DNA extraction techniques namely salting-out and classic phenol-chloroform. MATERIALS AND METHODS Qualification of two different DNA extraction techniques for 634 clinical specimens highly suspe...
متن کاملSurface tension measurements show that chaotropic salting-in denaturants are not just water-structure breakers.
Since the salting-in agents guanidinium chloride, urea, and lithium perchlorate increase the surface tension of water, the salting-in phenomenon does not reflect easier cavity formation in water. Therefore, these salting-in agents must be directly contributing to the solvation of a solute such as benzene in water, probably by a direct solvation interaction. The increased surface-tension effects...
متن کاملComparison of Three Different DNA Extraction Methods for Linguatula serrata as a Food Born Pathogen
BACKGROUND One of the most important items in molecular characterization of food-borne pathogens is high quality genomic DNA. In this study, we investigated three protocols and compared their simplicity, duration and costs for extracting genomic DNA from Linguatula serrata. METHODS The larvae were collected from the sheep's visceral organs from the Yazd Slaughterhouse during May 2013. DNA ext...
متن کامل1H NMR and molecular dynamics evidence for an unexpected interaction on the origin of salting-in/salting-out phenomena.
By employing (1)H NMR spectroscopy and molecular simulations, we provide an explanation for recent observations that the aqueous solubilities of ionic liquids exhibit salting-out to salting-in regimes upon addition of distinct inorganic salt ions. Using a typical ionic liquid [1-butyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide], we observed the existence of preferential specific inte...
